The prepared cationic albumin proteins interact with anionic albumin proteins to form albumin PEC NPs (110 nm) with +37 mV surface zeta potential. Furthermore, albumin PEC NPs preparation in water alone achieves chlorambucil (CHL) loading up to 17 times higher than the conventional desolvation method, largely due to the elimination of drug loss to organic solvents. CHL loaded albumin PEC NPs also decreased the cell viability (Huh-7) to 44 % within 24 h. This study demonstrates that high drug-loaded albumin NPs can be alternatively synthesized by using albumin polyelectrolyte properties, and applied in drug delivery applications. © 2025 Elsevier B.V., All rights reserved.eninfo:eu-repo/semantics/closedAccessAlbumin PolyelectrolyteAqueous MediaCell ViabilityDrug DeliveryNanoparticleChlorambucilWaterAlbuminsChlorambucilDrug CarriersPolyelectrolytesSerum Albumin, BovineWaterBiotek Synergy H1 Microplate ReaderGraphpad Prism 10.0ImageJQuanta 250 FEGZeiss LSM 880Controlled Drug DeliveryDyesOrganic SolventsProteinsTargeted Drug DeliveryAlbumin NanoparticlesAlbumin PolyelectrolyteAqueous MediaCell ViabilityComplex FormationsComplex NanoparticlesHigh Drug LoadingsOrganicsPolyelectrolyte ComplexesNanoparticlesAlbuminBovine Serum AlbuminChlorambucilNanoparticlePolyelectrolyteWaterDrug CarrierAqueous SolutionArticleCell ViabilityCentrifugationConfocal MicroscopyControlled StudyDrug Delivery SystemEnzymatic DegradationHEK293T Cell LineHuh-7 Cell LineHumanHuman CellIncubation TimeMCF-7 Cell LineParticle SizePhoton Correlation SpectroscopyTurbidityZeta PotentialAnimalCell SurvivalChemistryDrug EffectGreen ChemistryTumor Cell LineUltrastructureAlbuminsAnimalsCell Line, TumorCell SurvivalChlorambucilDrug CarriersDrug Delivery SystemsGreen Chemistry TechnologyHumansPolyelectrolytesSerum Albumin, BovineWaterA Green Route To Albumin/Albumin Polyelectrolyte Complex Nanoparticles in Water With High Drug Loading for Drug DeliveryArticle2-s2.0-10501321266710.1016/j.ijbiomac.2025.146978
